Effect of Dicyclohexylcarbodiimide on the Microsomal ATPase Activity of the Rat Kidney

Abstract
The present report was an attempt to study the effect of dicyclohexylcarbodiimide (DCCD) on both Mg++ and Na+-K+ activated ATPases of microsomal fraction obtained from rat kidney in vitro under the¡©variety of conditions. The results were summarized as follows.
1. The activities of Mg++ ATPase and Na+-K+ ATPase are both depressed markedly at the concentration. of 0.2 mM DCCD.
2. The inhibition of Na+-K+ ATPase activity. of microsomal fraction of the kidney by DCCD increases in lower pH and in the addition of hydroxylamine.
3. N-ethylmaleimide; depresses markedly Na+-K+ ATPase, and the inhibition of Na+-K+ ATPase activity by DCCD is not prevented by an addition of cysteine in the medium.
These findings indicate that Na+-K+ ATPase system isolated from microsomal fraction¢¥ of the rat kidney is quite similar to that from other tissues such as the brain and the erythrocytes, and remarkable corre¡©lation between action of Na+-K+ ATPase and sulphydryl or carboxyl groups could be observed.
